Description Cytogenetic analysis of chorionic villus biopsy for prenatal diagnosis of chromosome abnormalities.

Preferred Collection Volume 25 mg
Minimum Collection Volume 15 mg
Collection Instructions Approximately 25 mg of unclean specimen usually provides enough material for cytogenetic studies.
Specimen Handling Place the chorionic villus sample into a tube containing 10 mL of sterile tissue culture medium.

Note: Deliver to lab at room temperature within 24 hours. If collected over a weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported Immediately. Do not freeze. Protect from heat or cold.
Rejection Criteria Frozen, fixed or grossly contaminated specimen.
Methodology Chorionic villi are cleaned by physical and enzymatic methods. Villi are minced, disaggregated, cultured in four to six vessels, and then harvested and G-banded with Trypsin-Wright stain. The chromosomes in twenty metaphase spreads from at least 2 separate cultures are counted. Five cells are fully analyzed, and two are karyotyped.
