Description Fluorescence in situ Hybridization (FISH) is performed to detect the MDM2 (12q15) gene amplification, most commonly observed in sarcomas of soft tissue.
Specimen Requirements 
Container Cytogenetics transport media (call Cytogenetics Laboratory to collect media tubes)
Specimen Type Tissue;tissue sections
Preferred Collection Volume 5mm3 tissue; 2 unstained positively charged slides cut at 4 microns and one H&E stain slide.

Specimen Handling
Rejection Criteria Frozen or fixed tissue specimens, or tissue specimens sent without tissue culture medium; cases submitted without H&E slide.
Methodology A dual color fluorescent labeled DNA probe for the MDM2 gene, at chromosome band 12q15 is hybridized to unstained interphase nuclei on a slide. 200 cells are evaluated to determine if there is an amplification.
