Description Fluorescence In Situ Hybridization (F SH) is performed to evaluate the number of signals for chromosomes 13, 18, 21, X and Y In each cell. Useful as a rapid screen for aneuploidy on blood from a critically ill newborn or tissue from fetal demise. Result must be confirmed by routine chromosome analysis
Specimen Requirements 
Container Dark green top (sodium heparin) collection tube; sterile tissue culture medium

Preferred Collection Volume 5-10 mL blood; 5mm³ tissue
Minimum Collection Volume 5 mL blood; 5mm³ tissue
Collection Instructions 1. Blood: 5-10 mL eseptically collected blood in a green top, sodium heparin tube (or 3 mL pediatric green top, sodium heparin tube). Do not use lithium heparin tubes.
2. Tissue: At least 5mm³ of aseptically collected tissue. Kidney, lung, fascia, cartilage and chorionic villi are preferred specimens from autopsy. Do not send skin from autopsy. Place specimen in 10 mL of sterile tissue culture medium and refrigerate until transported. Do not freeze. Do not put in formalin.
Specimen Handling Deliver to lab at room temperature within 24 hours. If collected over the weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported immediately. Do not freeze. Protect from heat or cold.
Rejection Criteria Frozen, clotted, hemolyzed or grossly contaminated specimens. Incorrect anticoagulant used. Frozen or fixed tissue specimens, or tissue specimens sent without tissue culture medium.
Methodology Fluorescent labeled DNA probes specific for the centromeres of chromosomes X (DXZ1), Y (DYZ3), and 18 (D18Z1), and for chromosome bands 13q14 (RB1) and 21q22.13-q22.2 (D21S259, D21S341, D21S342) are hybridized to unstained interphase nuclei on a slide. 100 cells are scored for the number of each fluorescent signal.
