Description Fluorescence In situ hybridization (FISH) Is performed to detect the (15;17) translocation that is diagnostic of acute promyelocytlc leukemia, FAB type M3.
Specimen Requirements

Specimen Handling Deliver to lab at room temperature within 24 hours. If collected over a weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported immediately. Do not freeze. Protect from heat or cold.
Rejection Criteria Frozen, clotted, hemolyzed or grossly contaminated specimens. Incorrect anticoagulant used.
Methodology Dual color fluorescent labeled DNA probes for the PML gene, at chromosome band 15q24, and the RARa gene, at chromosome band 17q21.1, are hybridized to unstained interphase nuclei on a slide. 200 cells are evaluated to determine if the signals are discreet (no translocation) or fused (translocation).
