Description Loss of heterozygosity (LOH) involving 1p and 19q is a common genetic alteration in oligodendroglioma (brain tumor). Positive 1p/19q LOH has been associated with chemosensitivity, extended response to chemotherapy, and extended survival.

Specimen Handling Please send either a paraffin block or two H&E recuts and 3 unstained paraffin sections from the same paraffin block. Preferably, the tissue should contain both areas of tumor and normal brain, but just tumor is acceptable as well. Include a copy of the pathology report. If a neuropathology consultation is requested in addition to a 1p/19q LOH test, send all microscopic slides, a representative paraffin block, brief clinical history, and preoperative radiology report or MRI scans on a CD.

Rejection Criteria
Methodology This assay is performed as described (Hatanpaa et al., Lab Invest 83. 419-428.2003 (see link below)) with small modifications. An area of tumor is identified on an H&E section by a neuropathologist. DNA is extracted from the corresponding area in an unstained paraffin section. Highly polymorphic dinucleotide repeat loci on 1p and 19q are amplified by PCR. Fragment analysis is performed by capillary electrophoresis.
